Polycaprolactone-Based Scaffolds Facilitates Osteogenic Differentiation of Human Adipose-Derived Stem Cells in a Co-Culture System

(1) Background: Stem cells in combination with scaffolds and bioactive molecules have made significant contributions to the regeneration of damaged bone tissues. A co-culture system can be effective enhancing proliferation rate osteogenic differentiation stem cells. Hence, aim this study was investigate human adipose derived when co-cultured osteoblasts seeded on polycaprolactone (PCL):hydroxyapatite (HA) scaffold; (2) Methods: Human adipose-derived (ASC) (HOB) were three different ratios 1:2, 1:2 2:1 PCL-HA scaffolds. The ability evaluated based cell morphology, rate, alkaline phosphatase (ALP) activity, calcium deposition genes expression levels using quantitative RT-PCR; (3) Results: ASC/HOB ratio showed most positive as compared other groups, which resulted higher ALP expression, particularly Runx, BSP. These indicate that at early stage development; (4) Conclusions: (ASC/HOB) promote formation.
